Immunohistochemistry

Assay Principle
AEC is a widely used chromogen for immunohistochemical staining. AEC produces a red end product that is soluble in alcohol and must be used with an aqueous counterstain and mounting media.
Assay Protocol
1. After the peroxidase detection, wash with TBS or PBS for 4 times, 5 minutes each
2. Add one drop of chromogenic reagent A and B respectively into 1 ml of distilled water and mix well. Add it onto the sample and colorate in dark for 10-30 minutes.
3. Wash the slide gently with distilled water to stop the reaction. Counterstain with methyl blue for 3-5 minutes or nonalcoholic hematoxylin (Mayer’s) if necessary.
4. Seal the slide with aqueous mounting medium (Cat # AR1018) or glycerol. Observe with microscope.
Note
Counterstain with alcoholic hematoxylin is not recommended.
