Background of ABCB11

Bile Salt Export Pump (BSEP) is a protein which in humans is encoded by the ABCB 11 gene. It is a member of the superfamily of ATP-binding cassette (ABC) transporters, also known as ABCB11. It is mapped to chromosome 2q24. The BSEP protein is mainly expressed in the liver. ABCB11 is a gene associated with progressive familial intrahepatic cholestasis type 2 (PFIC2) which caused by mutations in the ABCB11 gene will increases the risk of hepatocellular carcinoma in early life.

Would PB9414 anti-ABCB11 antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?

Answer

You can see on the product datasheet, PB9414 anti-ABCB11 antibody as been tested on ICC. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.

We are currently using anti-ABCB11 antibody PB9414 for human tissue, and we are well pleased with the ICC results. The species of reactivity given in the datasheet says human, mouse, rat. Is it true that the antibody can work on canine tissues as well?
