Protein Target Info (Source: Uniprot.org)
You can check the tissue specificity below for information on selecting positive and negative control.
|Protein Name||Angiotensin-converting enzyme|
|Molecular Weight||149715 MW|
|Protein Function||Converts angiotensin I to angiotensin II by release of the terminal His-Leu, this results in an increase of the vasoconstrictor activity of angiotensin. Also able to inactivate bradykinin, a potent vasodilator. Has also a glycosidase activity which releases GPI-anchored proteins from the membrane by cleaving the mannose linkage in the GPI moiety.|
|Tissue Specificity||Ubiquitously expressed, with highest levels in lung, kidney, heart, gastrointestinal system and prostate. Isoform Testis-specific is expressed in spermatocytes and adult testis. .|
|Sequence Similarities||Belongs to the peptidase M2 family.|
|Subcellular Localization||Angiotensin-converting enzyme, soluble form: Secreted.|
|Alternative Names||Angiotensin-converting enzyme;ACE;3.2.1.-;126.96.36.199;Dipeptidyl carboxypeptidase I;Kininase II;CD143;Angiotensin-converting enzyme, soluble form;ACE;DCP, DCP1;|

Predicted bind size: 150KD
Observed bind size: 150KD
1. Post-translational modification:phosphorylation, methylation, glycosylation etc. These modifications prevent SDS molecules from binding to the target protein and thus make the band size appear larger than expected
2. Post-translational cleavage: this can cause smaller bands and or multiple bands
3. Alternative splicing: the same gene can have alternative splicing patterns generating different size proteins, all with reactivities to the antibody.
4. Amino Acid R chain charge: SDS binds to positive charges. The different size and charge of the Amino Acid side chains can affect the amount of SDS binding and thus affect the observed band size.
5. Multimers: Multimers are usually broken up in reducing conditions. However if the interactions between the multimers are strong, the band may appear higher.,
