Background of FBXW11
This gene encodes a member of the F-box protein family which is characterized by an approximately 40 amino acid motif, the F-box. The F-box proteins constitute one of the four subunits of ubiquitin protein ligase complex called SCFs (SKP1-cullin-F-box), which function in phosphorylation-dependent ubiquitination. The F-box proteins are divided into 3 classes: Fbws containing WD-40 domains, Fbls containing leucine-rich repeats, and Fbxs containing either different protein-protein interaction modules or no recognizable motifs. The protein encoded by this gene belongs to the Fbws class and, in addition to an F-box, contains multiple WD40 repeats. This gene contains at least 14 exons, and its alternative splicing generates 3 transcript variants diverging at the presence/absence of two alternate exons.

Western blot analysis of Beta TrCP2 expression in NIH3T3 whole cell lysates (lane 1) and 293 whole cell lysates (lane 2). Beta TrCP2 at 58 kDa was detected using rabbit anti-Beta TrCP2 affinity purified polyclonal antibody (Catalog # A05523) at 1:200. The blot was developed using chemiluminescence (ECL) method (Catalog # EK1002).

Question
Can you help my question with product A05523, anti-Beta TrCP2 antibody. I was wondering if it would be possible to conjugate this antibody with biotin. I would need it to be without BSA or sodium azide. I am planning on using a buffer exchange of sodium azide with PBS only. Would there be problems for me to conjugate the antibody and store it in -20 degrees in small aliquots?

Answer
We suggest not storing this antibody with PBS buffer only in -20 degrees. If you want to store it in -20 degrees it is best to add some cryoprotectant like glycerol. If you want carrier free A05523 anti-Beta TrCP2 antibody, we can provide it to you in a special formula with trehalose and/or glycerol. These molecules will not interfere with conjugation chemistry and provide a good level of protection for the antibody from degradation. Please be sure to specify this in your purchase order.

We are currently using anti-Beta TrCP2 antibody A05523 for human tissue, and we are content with the WB results. The species of reactivity given in the datasheet says human. Is it true that the antibody can work on goat tissues as well?

The anti-Beta TrCP2 antibody (A05523) has not been validated for cross reactivity specifically with goat tissues, though there is a good chance of cross reactivity.
