Background of Calponin 1

Multiple isoelectric variants of calponin have been identified, however only two molecular weight isoforms exist; a 34kDa form and a 29kDa form. Expression of the 29kDa form, I-calponin, is primarily restricted to muscle of the urogenital tract, whereas the higher molecular weight variant has been demonstrated in vascular and visceral smooth muscle. In Western blotting, This monoclonal antibody reacts with only the 34kDa form of calponin in extracts of human aortic medial smooth muscle and is unreactive with fibroblast extracts of cultivated human foreskin. Calponin is a calmodulin, F-actin and tropomyosin binding protein, which is thought to be involved in the regulation of smooth muscle contraction. Calponin expression is restricted to smooth muscle cells and has been shown to be a marker of the differentiated (contractile) phenotype of developing smooth muscle.

Antibody Validation

Boster validates all antibodies on WB, IHC, ICC, Immunofluorescence, and ELISA with known positive control and negative samples to ensure specificity and high affinity, including thorough antibody incubations.

Assay Dilutions Recommendation

The recommendations below provide a starting point for assay optimization. The actual working concentration varies and should be decided by the user.

Western Blot (1-2ug/ml)
Flow Cytometry (1-2ug/million cells)
Immunohistochemistry (Formalin-fixed) (1-2ug/ml for 30 minutes at RT)(Staining of formalin-fixed tissues requires heating tissue sections in 1mM EDTA, pH 7.5-8.5, for 45 min at 95°C followed by cooling at RT for 20 minutes)Optimal dilution for a specific application should be determined.

3 Customer Q&As for Anti-Calponin-1 (Smooth Muscle Marker) Monoclonal Antibody

Question

Would anti-Calponin-1 (Smooth Muscle Marker) Monoclonal antibody M08065 work on feline IHC with pericardium small intestine?

Verified Customer

Verified customer

Asked: 2018-07-02

Answer

Our lab technicians have not tested anti-Calponin-1 (Smooth Muscle Marker) Monoclonal antibody M08065 on feline. You can run a BLAST between feline and the immunogen sequence of anti-Calponin-1 (Smooth Muscle Marker) Monoclonal antibody M08065 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated feline samples, this use of the antibody is not covered by our guarantee. However we have an innovator award program that if you test this antibody and show it works in feline pericardium small intestine in IHC, you can get your next antibody for free.

Question

Would M08065 anti-Calponin-1 (Smooth Muscle Marker) Monoclonal antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?

T. Li

Verified customer

Asked: 2013-12-18

Answer

You can see on the product datasheet, M08065 anti-Calponin-1 (Smooth Muscle Marker) Monoclonal antibody as been validated on IHC. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.
