|Molecular Weight||18502 MW|
|Protein Function||Binds to F-actin and exhibits pH-sensitive F-actin depolymerizing activity. Regulates actin cytoskeleton dynamics. Important for normal progress through mitosis and normal cytokinesis. Plays a role in the regulation of cell morphology and cytoskeletal organization. Required for the up-regulation of atypical chemokine receptor ACKR2 from endosomal compartment to cell membrane, increasing its efficiency in chemokine uptake and degradation. .|
|Tissue Specificity||Widely distributed in various tissues.|
|Sequence Similarities||Belongs to the actin-binding proteins ADF family.|
|Subcellular Localization||Nucleus matrix . Cytoplasm, cytoskeleton . Cell projection, ruffle membrane ; Peripheral membrane protein ; Cytoplasmic side . Cell projection, lamellipodium membrane ; Peripheral membrane protein ; Cytoplasmic side . Colocalizes with the actin cytoskeleton in membrane ruffles and lamellipodia. Detected at the cleavage furrow and contractile ring during cytokinesis. Almost completely in nucleus in cells exposed to heat shock or 10% dimethyl sulfoxide.|

All lanes: Anti-Cofilin(PB9033) at 0.5ug/ml
WB: Recombinant Human Cofilin Protein 0.5ng
Predicted bind size: 36KD
Observed bind size: 36KD
1. Post-translational modification:phosphorylation, methylation, glycosylation etc. These modifications prevent SDS molecules from binding to the target protein and thus make the band size appear larger than expected
2. Post-translational cleavage: this can cause smaller bands and or multiple bands
3. Alternative splicing: the same gene can have alternative splicing patterns generating different size proteins, all with reactivities to the antibody.
4. Amino Acid R chain charge: SDS binds to positive charges. The different size and charge of the Amino Acid side chains can affect the amount of SDS binding and thus affect the observed band size.
5. Multimers: Multimers are usually broken up in reducing conditions. However if the interactions between the multimers are strong, the band may appear higher.,
