Background of GluR5/GRIK1

Glutamate receptor, ionotropic, kainate 1, also known as GLUR5, is a protein that in humans is encoded by the GRIK1 gene. It is mapped to 21q21.3. This gene encodes one of the many ionotropic glutamate receptor (GluR) subunits that function as a ligand-gated ion channel. The specific GluR subunit encoded by this gene is of the kainate receptor subtype. Receptor assembly and intracellular trafficking of ionotropic glutamate receptors are regulated by RNA editing and alternative splicing. These receptors mediate excitatory neurotransmission and are critical for normal synaptic function. Exons of this gene are interspersed with exons from the C21or f41 gene, which is transcribed in the same orientation as this gene but does not seem to encode a protein.

Antibody Validation

Boster validates all antibodies on WB, IHC, ICC, Immunofluorescence, and ELISA with known positive control and negative samples to ensure specificity and high affinity, including thorough antibody incubations.

Question

Would PB9208 anti-GRIK1 antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?

Verified Customer

Verified customer

Asked: 2020-03-30

Answer

As indicated on the product datasheet, PB9208 anti-GRIK1 antibody as been tested on IHC-P. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.

Question

Is this PB9208 anti-GRIK1 antibody reactive to the isotypes of GRIK1?

Verified Customer

Verified customer

Asked: 2019-09-03

Answer

The immunogen of PB9208 anti-GRIK1 antibody is E.coli-derived human GRIK1 recombinant protein (Position: R271-I450). Human GRIK1 shares 88% and 94% amino acid (aa) sequences identity with mouse and rat GRIK1, respectively. Could you tell me which isotype you are interested in so I can help see if the immunogen is part of this isotype?

Question

We are currently using anti-GRIK1 antibody PB9208 for mouse tissue, and we are well pleased with the IHC-P results. The species of reactivity given in the datasheet says human, mouse, rat. Is it possible that the antibody can work on primate tissues as well?

D. Patel

Verified customer

Asked: 2019-07-11

Answer

The anti-GRIK1 antibody (PB9208) has not been validated for cross reactivity specifically with primate tissues, though there is a good chance of cross reactivity. We have an innovator award program that if you test this antibody and show it works in primate you can get your next antibody for free. Please contact me if I can help you with anything.
