Background for Insulin-like growth factor I(IGF-I)

Insulin-like growth factor 1(IGF-1) also known as somatomedin C or mechano growth factor is a protein that in humans is encoded by the IGF1 gene. IGF-1 is a hormone similar in molecular structure to insulin. It plays an important role in childhood growth and continues to have anabolic effects in adults. A synthetic analog of IGF-1, mecasermin is used for the treatment of growth failure. IGF-1 consists of 70 amino acids in a single chain with three intramolecular disulfide bridges. IGF-1 has a molecular weight of 7649 daltons.Justice et al.(1990) placed the mouse IGF1 gene on chromosome 10.

Q: How should I reconstitute my WB control (IGF-1 recombinant protein 100 ng/vial) for working with the mouse muscle tissue? I have tried a couple of different dilutions already. It appeared that much of the protein stayed lodged at the top of the gel and did not migrate (Evidence from the signal saturation at the top of the blot in the control lane).
A: We suggest you use 50 µL water to reconstitute with the 100 ng recombinant protein. Then mix 10 µL of this solution with 10 µL of 2X sample buffer so that the protein concentration is 1 ng/µl (i.e., 1 µg/mL).
Q: I did not see a band in my samples at 8 kDa. Instead, I saw a very well-defined clear band at about 12.5 kDa. The positive control had a signal at 10 kDa. Does the mouse IGF-1 run higher than the human IGF-1? I used a 15% acrylamide gel.
A; There might be problem that your pre-stained markers did not give the required precision on your gel. Note that the nominal molecular weight of mouse IGF-1 is different from that of human.

Q: What is the expected band size? Why is it different than the observed band size?
A: The expected band size is predicted on the size of the protein. The actual band size may be affected by a few other factors including but not limited to:
1. Post-translational modification:phosphorylation, methylation, glycosylation etc. These modifications prevent SDS molecules from binding to the target protein and thus make the band size appear larger than expected
2. Post-translational cleavage: this can cause smaller bands and or multiple bands

3. Alternative splicing: the same gene can have alternative splicing patterns generating different size proteins, all with reactivities to the antibody.

4. Amino Acid R chain charge: SDS binds to positive charges. The different size and charge of the Amino Acid side chains can affect the amount of SDS binding and thus affect the observed band size.
5. Multimers: Multimers are usually broken up in reducing conditions. However if the interactions between the multimers are strong, the band may appear higher.,
