Background of MAG/Siglec-4a

MAG (Myelin-associated glycoprotein),also known as SIGLEC4A, is a cell membrane glycoprotein that is a member of the SIGLEC family of proteins and is a functional ligand of the NOGO-66 receptor, NgR. It is though to be involved in the process of myelination. MAG is a sialic acid-binding SIGLEC protein and is a functional ligand for the NOGO receptor. The MAG gene is mapped on 19q13.12. Cleavage of GPI-linked proteins from axons protects growth cones from MAG-induced collapse, and dominant-negative NgR eliminates MAG inhibition of neurite outgrowth. MAG-resistant embryonic neurons were rendered MAG-sensitive by expression of NgR. MAG binds specifically to an NgR-expressing cell line in a GPI-dependent and sialic acid-independent manner. Experiments blocking NgR from interacting with MAG prevented inhibition of neurite outgrowth by MAG. In cultured human embryonic kidney (HEK) cells expressing the NOGO receptor, p75 (NTR) was required for MAG-induced intracellular calcium elevation.

Question

Will PA1751 anti-MAG antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?

Verified Customer

Verified customer

Asked: 2019-12-16

Answer

As indicated on the product datasheet, PA1751 anti-MAG antibody as been tested on IHC. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.

Answer

Our lab technicians have not validated anti-MAG antibody PA1751 on bovine. You can run a BLAST between bovine and the immunogen sequence of anti-MAG antibody PA1751 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated bovine samples, this use of the antibody is not covered by our guarantee. Question

Dear Boster: I have some few question about your product Anti-MAG Antibody (PA1751). Would this antibody recognize both S and L MAG? Would it be appropriate to use on mouse CNS tissue for WB? Would it recognize deglycosylated forms of MAG?

Verified Customer

Verified customer

Asked: 2017-06-19

Answer

The product Anti-MAG Antibody (PA1751) have been tested and validated to work with mouse samples in Western Blot. Unfortunately, we do not have any data whether this antibody recognize S- or/and L-MAG as this has not been tested. Also we do not have any information about the difference between S- and L-MAG, but as the theory suggest that if it contain the sequence used to generate the antibody, then it should be able to be detected, again we cannot guarantee as it has not been tested the same also for deglycosylated MAG.

Question

Can you help my question with product PA1751, anti-MAG antibody. I was wondering if it would be possible to conjugate this antibody with biotin. I would need it to be without BSA or sodium azide. I am planning on using a buffer exchange of sodium azide with PBS only. Would there be problems for me to conjugate the antibody and store it in -20 degrees in small aliquots?

B. Li

Verified customer

Asked: 2016-08-04

Answer

We do not recommend storing this antibody with PBS buffer only in -20 degrees. If you want to store it in -20 degrees it is best to add some cryoprotectant like glycerol. If you want carrier free PA1751 anti-MAG antibody, we can provide it to you in a special formula with trehalose and/or glycerol. These molecules will not interfere with conjugation chemistry and provide a good level of protection for the antibody from degradation. Please be sure to specify this in your purchase order.
