Protein Function Promotes microtubule assembly and stability, and might be involved in the establishment and maintenance of neuronal polarity. The C-terminus binds axonal microtubules while the N- terminus binds neural plasma membrane components, suggesting that tau functions as a linker protein between both. Axonal polarity is predetermined by tau localization (in the neuronal cell) in the domain of the cell body defined by the centrosome. The short isoforms allow plasticity of the cytoskeleton whereas the longer isoforms may preferentially play a role in its stabilization.
Tissue Specificity Expressed in neurons. The larger forms (isoform tau-A and isoform tau-B) are preferentially expressed in the peripheral nervous system while the other are expressed in the central nervous system. Low amounts of the larger forms are also found in limited areas of the CNS.

Background for Microtubule-associated protein tau

The microtubule-associated proteins(MAPs) coassemble with tubulin into microtubules in vitro. Microtubule-associated protein tau appears to be enriched in axons. Tau are composed of 352 to 441 amino acids. The isoforms differ from each other by the presence or absence of 29-amino acid or 58-amino acid inserts located in the N terminus and a 31-amino repeat located in the C terminus. tau is important in establishing and maintaining neuronal morphology and is a major component of the neurofibrillary tangles(NFTs) characteristic of Alzheimer's brain. Microtubule-associated protein tau(MTBT1) is mapped to chromosome 17q21.
