Background of TRAF-4
F-actin cross-linking protein which is thought to anchor actin to a variety of intracellular structures. This is a bundling protein. Probably involved in vesicular trafficking via its association with the CART complex. The CART complex is necessary for efficient transferrin receptor recycling but not for EGFR degradation.

Why can't we get a signal when using M03069 on HELA and liver tissue WB?

Please see the following tips to improve test result using the Anti-TRAF4 Rabbit Monoclonal Antibody (M03069): 1. Please use lysis buffer with 1%SDS if the LDS lysis buffer is a non-reducing buffer. 2. The membrane cutting area should preferably be within 20-80kDa. Place 50kDa in the middle of PVDF membrane. 3. Load equal amounts (20-30 ug) of protein into each well of the SDS-PAGE gel, along with appropriate molecular weight marker. 1 ug is too little. 4. Block the membrane for 1 hour at room temperature using 1%BSA/TBST or 2-3% non-fat Milk/ TBST. Dilute the antibodies for 1 hour at room temperature using 1%BSA/TBST or 2-3% non-fat Milk/ TBST. 5. Increase the concentration of the primary antibody and the secondary antibody.e.g. primary antibody 1:500-1000, secondary antibody 1:1000-2000. 6. Do not overuse ECL substrate. After substrate incubation, remove the excess ECL substrate from the membrane before exposure (place the membrane vertically, and the ECL substrate liquid does not flow down, and then use paper to absorb the excess ECL substrate around the membrane). This can prevent the flow of ECL substrate from causing strong non-specific bands, dark background and irregular black marks.
