Boster’s BCIP/ NBT Chromogenic Substrate Kit are ready-made solutions for chromogenic blot and immunohistochemical staining applications. NBT nitro-blue tetrazolium and BCIP 5-bromo-4-chloro-3'-indolyphosphate results in an intense, black purple precipitate after reacting with alkaline phosphatase.
1. After the alkaline phosphatase detection, wash fully with TBS 4 times and each time for 5 minutes. Do not use phosphate buffer.
2. Add one drop of chromogenic reagent A and B respectively into 1ml of distilled water and mix evenly. Add it to the sample and colorate in dark for 10-30 minutes. If no background appearing, keep coloration until satisfied.
3. Wash fully with distilled water to stop reaction. Counterstain with nuclear fast red if necessary.
4. Seal sllide with Aqueous Mounting Medium (AR1018). Observe with microscope
