Background of BAFF/BLyS/TNFSF13B

B-cell activating factor (BAFF) also known as tumor necrosis factor ligand superfamily member 13B is a protein that in humans is encoded by the TNFLSF13B gene.1,2 BAFF is a cytokine that belongs to the tumor necrosis factor (TNF) ligand family. This cytokine is a ligand for receptors TNFRSF13B/TACI, TNFRSF17/BCMA, and TNFRSF13C/BAFFR. This cytokine is expressed in B cell lineage cells, and acts as a potent B cell activator. It has been also shown to play an important role in the proliferation and differentiation of B cells.3All these ligands act as heterotrimers (i.e. three of the same molecule) interacting with heterotrimeric receptors4, although BAFF has been known to be active as either a hetero- or homotrimer.5 The standard used in this kit is recombinant mouse soluble BAFF (A127-L309) with molecular weight 23.2KDa.

Question

Q: how much samples can be assayed in a Picokine® ELISA Kit?

K. Gonzalez

Verified customer

Asked: 2019-03-20

Answer

A: The Picokine® ELISA Kits will generally run a 7-point standard curve, non-specific binding wells, and 39 samples in duplicate. this may might differ by kit so please refer to each datasheet for details.

Question

Q: how long can samples (cell cultures, serum, and plasma) be stored and still be stable for analysis BAFF using the EK0664 Mouse BAFF Picokine® ELISA Kit?

Verified Customer

Verified customer

Asked: 2019-01-02

Answer

A: Boster Bio does not evaluate sample stability. Variations in sample collection, processing, and storage may change the stabilityof samples. It is recommend to assay sample right after collection when possible, or aliquot into single use volumes and store samples frozen. avoid repeat freeze-thaw cycles with the stored samples to avoid protein degradation.

Question

Q: how are cell lysates prepared for use in Picokine® ELISA kits?

Verified Customer

Verified customer

Asked: 2018-07-12

Answer

A: for those Picokine® ELISAs where cell or tissue lysate is a validated sample type, sample preparation instructions for lysate are present in the product insert. Components in lysate and lysis buffer can effect immunoreactivity, so if lysate is not a validated sample type, care must be taken in sample preparation and validation.

Question

Q: how can I thaw whole blood sample for BAFF ELISA after freezing?

Verified Customer

Verified customer

Asked: 2017-04-15

Answer

A: we do not recommend freezing and thaw whole blood. erythrocytes are fragile and, if frozen and thawed, will undergo hemolysis rendering the samples useless. To keep your blood samples to test BAFF for a later time, you should let the blood clot in glass tubes and separate the serum to freeze for later analysis.
