Phosphate buffered saline is widely applied to biological research such as washing cells and dilutions.

Usage and Handling

Background

Phosphate Buffered Saline (PBS) is an isotonic solution containing 0.15M sodium chloride and 0.02M phosphate in the form of disodium phosphate and monosodium phosphate. PBS is used to provide a buffer system to maintain physiological pH and osmolarity, essential for tissue cultures and molecular biology experiments. PBS can be utilized as a diluting agent in preparing decimal dilutions of active reagents, as a rinsing agent for rinsing cells and cell containers, as a media for samples preparation and transportation. It is a commonly used wash buffer in immunoassay procedures, reducing non-specific binding by effectively removing unbound reagents thus preventing high background and improving the sensitivity of the assay.

Phosphate Buffered Saline (PBS) Recipe

Below is the phosphate-buffered saline recipe. The required reagents are required for formulations of :

• 800 g NaCl

• 20 g KCl

• 144 g Na2HPO4 · 2H2O

• 24 g KH2PO4

• 8 L of distilled water l

Take the following steps in PBS protocol to make a stock solution of Phosphate buffered saline (PBS)

1. Be ready with 800 mL of distilled water in a convenient container.

2. Add 8 g of NaCl to the solution.

3. Add 200 mg of KCl to the solution.

4. Add 1.44 g of Na2HPO4 to the solution.

5. Add 240 mg of KH2PO4 to the solution.

6. Adjust solution to desired pH (typically pH ≈ 7.4).

7. Add distilled water until volume is 1 L.

Take the stock solution and dispense the phosphate buffered saline (PBS) into aliquots and sterilize through autoclaving for 20 minutes for 121 degrees celsius.
