Trypan Blue is a vital cell stain used to assess cell viability using the dye exclusion test. Live cells with intact cell membranes are not coloured. Since cells are very selective in the compounds that pass through the membrane, in a viable cell trypan blue is not absorbed; however, it traverses the membrane in a dead cell. Hence, dead cells are shown as a distinctive blue colour under a microscope. And Trypan blue is commonly used for cell counting. The method cannot distinguish between necrotic and apoptotic cells.
1. Cell Collection
a. Trypsinize adherent cells or digest cells with EDTA; For suspending cells, collect directly.
b. Centrifuge at ~1000-2000 x g for 1 min, remove supernatant.
c. Add some cell diluent buffer into the sediment and resuspend pellet.
a. Add 90 μl cell suspension into EP tube.
b. Add 10 μl trypan blue solution (2X) in the EP tube and mix thoroughly.
c. Place in a CO2 incubator for 3-5 min. Note: Do not incubate too long.
3. Cell Viability Analysis
Place the cell suspension with trypan blue solution onto blood cell counting plate. Calculate the total number of cells and the number of cells dyed to blue respectively.
Cell Viability = (total number of cells - number of blue cells) / total number of cells X 100%
