1. Antigen Preparation and Immunization

Your project begins with antigen validation and immunization schedule selection. We require 4 mg of purified peptide or 2 mg of purified protein to start. Choose between our short immunization program for broader epitope coverage or long immunization program for higher affinity antibodies. Our team uses proven immune-stimulating protocols optimized for your specific antigen type.

2. Fusion and Hybridoma Generation

Following successful immunization, spleen and lymph node cells from immunized mice are fused with myeloma cells to create hybridoma cell lines. Our optimized fusion protocol generates approximately 192 oligoclones per project, each cultivated under selective conditions to ensure stable antibody-producing cell lines.

3. Screening and Selection

All hybridoma supernatants are screened by ELISA to identify positive clones. We then offer application-specific screening options including Western blot, immunofluorescence, flow cytometry, or custom assays in your lab. This ensures the selected antibodies perform exactly as needed for your downstream applications.

4. Subcloning to Monoclonality

Selected positive clones undergo limiting dilution subcloning to achieve true monoclonal stability. Multiple rounds of subcloning ensure genetic homogeneity, and each clone is validated through your chosen screening method. The final monoclonal hybridoma is then expanded for antibody production.

5. Production and Quality Control

Validated hybridoma clones are used to produce monoclonal antibodies using the ascites method. Each lot undergoes rigorous quality control testing including titer determination, specificity validation, and application testing. Comprehensive QC data is provided for your records.

6. Delivery and Long-term Storage

Your custom monoclonal antibodies are delivered with full documentation, including QC data, screening results, and culture protocols. Hybridoma cells are cryopreserved for up to 12 months at no additional cost, with extended storage available if needed.
