Why does apelin run smaller than its predicted 8.6 kDa mass?
Preproapelin is cleaved at the signal peptide (residues 1-22) and propeptide (23-41) before secretion, and cleavage on pairs of basic residues generates shorter bioactive fragments. The mature secreted peptide detected on blots is therefore smaller than the 8.6 kDa precursor; expect bands near or below the dye front unless a high-percentage or Tricine gel is used.
Does apelin expression change across lactation stages?
Yes; apelin is abundantly secreted in colostrum but drops to lower levels in milk and decreases rapidly within days after parturition, remaining detectable even in commercial milk. When probing mammary secretions or milk-derived samples, match sample collection timing to the biological question, since band intensity will vary sharply with lactation stage rather than reflecting assay inconsistency.
Why might milk-based blocking buffers increase background?
Non-fat dry milk can contain endogenous apelin, since the protein is naturally abundant in colostrum and still present in milk. Using milk as a blocking agent risks introducing native apelin onto the membrane, producing false-positive or elevated background signal. BSA-based blocking is a safer choice for this secreted hormone to avoid confounding the true sample signal.
What transfer method to use for APLN Western blot?
Given the roughly 8.6 kDa mature peptide size, use a high-percentage or Tricine gel with a low-MW-optimized PVDF membrane (0.2 micron pore) and a short, low-voltage semi-dry or wet transfer to prevent blot-through. Apelin lacks disulfide bonds, so no reducing-agent-related transfer complications are expected, but brief transfer times help retain the small peptide on the membrane.
How should secreted apelin be quantified reliably?
Because apelin is secreted into extracellular space rather than retained intracellularly, standard intracellular loading controls are unsuitable for normalizing conditioned media, serum, or milk samples. Quantify using a fixed loading volume, a secreted-protein or total-protein stain for normalization, and parallel standards of known apelin peptide concentration rather than relying on housekeeping gene products.
Why do multiple low-molecular-weight bands appear?
Preproapelin undergoes cleavage at paired basic residues to yield several bioactive fragments of differing length from a single precursor. Detecting multiple closely spaced, low-molecular-weight bands is consistent with partial or differential proteolytic processing rather than nonspecific binding, especially since the protein has no glycosylation or disulfide bonds to otherwise explain such shifts.
