Background of Frequenin

Frequenin, or Neuronal calcium sensor, is a regulator of G protein-coupled receptor phosphorylation in a calcium dependent manner. It directly regulates GRK1 (RHOK), but not GRK2 to GRK5. It can substitute for calmodulin and it stimulates PI4KB kinase activity. It is involved in long-term synaptic plasticity through its interaction with PICK1. Ncs1 may also play a role in neuron differentiation through inhibition of the activity of N-type voltage-gated calcium channel.

Antibody Validation

Boster validates all antibodies on WB, IHC, ICC, Immunofluorescence, and ELISA with known positive control and negative samples to ensure specificity and high affinity, including thorough antibody incubations.

Question

We are currently using anti-Frequenin antibody A05330 for rat tissue, and we are satisfied with the IF results. The species of reactivity given in the datasheet says human, mouse, rat. Is it likely that the antibody can work on zebrafish tissues as well?

Verified Customer

Verified customer

Asked: 2019-07-29

Answer

The anti-Frequenin antibody (A05330) has not been tested for cross reactivity specifically with zebrafish tissues, but there is a good chance of cross reactivity. We have an innovator award program that if you test this antibody and show it works in zebrafish you can get your next antibody for free. Please contact me if I can help you with anything.

Question

Would A05330 anti-Frequenin antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?

Verified Customer

Verified customer

Asked: 2019-05-20

Answer

You can see on the product datasheet, A05330 anti-Frequenin antibody as been tested on WB. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.

Question

Does anti-Frequenin antibody A05330 work on canine IF with dorsolateral prefrontal cortex?

Verified Customer

Verified customer

Asked: 2018-07-03

Answer

Our lab technicians have not validated anti-Frequenin antibody A05330 on canine. You can run a BLAST between canine and the immunogen sequence of anti-Frequenin antibody A05330 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated canine samples, this use of the antibody is not covered by our guarantee. However we have an innovator award program that if you test this antibody and show it works in canine dorsolateral prefrontal cortex in IF, you can get your next antibody for free.
