Which ALB phosphorylation sites matter when investigating multiple bands?
PTM · UniProt lists FAM20C phosphorylation at Ser29, Ser82, Ser89, and Thr107, plus phosphorylation at Ser297, Ser443, Thr444, Thr446, and Ser513. These are canonical precursor coordinates; antibody or paper numbering may differ. Their presence does not establish a visible shift. Compare samples under consistent preparation conditions before assigning a band to phosphorylation.

UniProt lists N-linked glycosylation at Asn342 in variant Redhill and Asn518 in variant Casebrook. These variant-specific sites should not be assumed present in every ALB sample. They are distinct from the listed lysine glycation sites. Confirm the variant before using either site to interpret a band.

ALB has a signal peptide at UniProt positions 1–18 and a propeptide at 19–24; processing removes these regions from mature secreted ALB. UniProt lists glycation at several lysines, including positions 36, 257, and 305; other sites, such as 75 and 161, are marked in vitro. Electrophoresis was performed on a 10% SDS-PAGE gel at 80V (Stacking gel) / 120V (Resolving gel) for 2 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions. <br>

Lane 2: rat liver tissue lysates,<br>
Lane 3: mouse blood lysates,<br>
Lane 4: mouse liver tissue lysates.<br>
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-Bovine Serum Albumin antigen affinity purified monoclonal antibody (M01245) at 1:500 overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:1000 for 1.5 hour at RT. The signal is developed using an ECL Plus Western Blotting Substrate (Catalog # AR1196-200) with Tanon 5200 system. A specific band was detected for Bovine Serum Albumin at approximately 69 kDa. The expected band size for Bovine Serum Albumin is at 69 kDa.
