FCGR2B were up-regulated in hippocampus of DM mice. A qRT-PCR was performed to detect the expression of ALB, AREG and FCGR2B mRNA expression in hippocampus of mice. B Western blot was conducted to detect the ALB, AREG and FCGR2B protein expression in hippocampus of mice. C IHC assay was employed to examine the ALB, AREG and FCGR2B protein expression in hippocampus of mice. D IF staining was utilized to detect the expression of FCGR2B and NeuN in hippocampus of mice. E Western blot was performed to detect the SHC1 protein expression in hippocampus of mice. F IF staining was performed to detect the expression of SHC1 and NeuN in hippocampus of mice. G Western blot was used to detect the p-PI3K and p-AKT protein expression in hippocampus of mice. *** P < 0.001 Full size image

Western blot analysis of Albumin using anti-Albumin antibody (M01245).
Electrophoresis was performed on a 10% SDS-PAGE gel at 80V (Stacking gel) / 120V (Resolving gel) for 2 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: rat blood lysates,
Lane 2: rat liver tissue lysates,
Lane 3: mouse blood lysates,
Lane 4: mouse liver tissue lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-Albumin antigen affinity purified monoclonal antibody (M01245) at 1:500 overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:1000 for 1.5 hour at RT. The signal is developed using an ECL Plus Western Blotting Substrate (Catalog # AR1196-200) with Tanon 5200 system. A specific band was detected for Albumin at approximately 69 kDa. The expected band size for Albumin is at 69 kDa.

| Results Summary | Human serum albumin (ALB) is the most abundant protein in plasma, with key functions including maintaining colloid osmotic pressure, transporting substances, providing antioxidant activity, and regulating inflammation. Clinically, ALB is widely used to treat hypoalbuminemia, shock, burns, and other conditions. ALB was long thought to be secreted exclusively by hepatocytes, but recent studies suggest that other cell types may also be capable of synthesizing it. The purpose of this experiment was to verify whether human skin keratinocytes can synthesize ALB. The results show that skin keratinocytes are indeed capable of producing ALB. |
