Flow Cytometry analysis of 293T cells using anti-YWHAH antibody (A04219-2).
Overlay histogram showing 293T cells stained with A04219-2(Blue line). The cells were fixed with 4% paraformaldehyde and blocked with 10% normal goat serum. And then incubated with rabbit anti-YWHAH Antibody (A04219-2, 1 μg/1x106 cells) for 30 min at 20°C. DyLight®488 conjugated goat anti-rabbit IgG (BA1127, 5-10 μg/1x106 cells) was used as secondary antibody for 30 minutes at 20°C. Isotype control antibody (Green line) was rabbit IgG (1 μg/1x106) used under the same conditions. Unlabelled sample without incubation with primary antibody and secondary antibody (Red line) was used as a blank control.

Suggested blocking solution with 5% non-fat milk or BSA; (*)Recommended protein loading: 20-40 ug per lane

Use TE buffer pH 9.0 for antigen retrieval; (*) citrate buffer pH 6.0 is an alternative.

Background of YWHAH
14-3-3 protein eta also referred to as 14-3-3η is a protein that in humans is encoded by the YWHAH gene. This gene product belongs to the 14-3-3 family of proteins which mediate signal transduction by binding to phosphoserine-containing proteins. This highly conserved protein family is found in both plants and mammals, and this protein is 99% identical to the mouse, rat and bovine orthologs. This gene contains a 7 bp repeat sequence in its 5' UTR, and changes in the number of this repeat have been associated with early-onset schizophrenia and psychotic bipolar disorder.
