Background of ANGPTL3
ANGPTL3 (Angiopoietin-Like 3), also known as ANGPT5, is a protein which in humans is encoded by the ANGPTL3 gene. The protein encoded by this gene is a member of the angiopoietin-like family of secreted factors. By radiation hybrid mapping and the use of surrounding genes, this gene is mapped to chromosome 1p31. It is predominantly expressed in the liver, and has the characteristic structure of angiopoietins, consisting of a signal peptide, N-terminal coiled-coil domain and the C-terminal fibrinogen (FBN)-like domain. Angptl3 also acts as dual inhibitor of lipoprotein lipase (LPL) and endothelial lipase (EL), and increases plasma triglyceride and HDL cholesterol in rodents. ANGPTL3 inhibit endothelial lipase to catalyze HDL-phospholipid and increase HDL-PL levels.

Western blot analysis of ANGPTL3 using anti-ANGPTL3 antibody (A02929).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
Lane 1: mouse HEPA1-6 cell lysate.
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-ANGPTL3 antigen affinity purified polyclonal antibody (Catalog # A02929) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for ANGPTL3 at approximately 54KD. The expected band size for ANGPTL3 is at 54KD.

Based on literature plasma does express ANGPTL3. Based on Uniprot.org, ANGPTL3 is expressed in liver, liver skeletal muscle, plasma, among other tissues. Regarding which tissues have ANGPTL3 expression, here are a few articles citing expression in various tissues:

Question
We are currently using anti-ANGPTL3 antibody A02929 for rat tissue, and we are well pleased with the WB results. The species of reactivity given in the datasheet says human, mouse, rat. Is it likely that the antibody can work on bovine tissues as well?

The anti-ANGPTL3 antibody (A02929) has not been validated for cross reactivity specifically with bovine tissues, but there is a good chance of cross reactivity. Our lab were happy with the WB result of your anti-ANGPTL3 antibody. However we have been able to see positive staining in liver secreted using this antibody. Is that expected? Could you tell me where is ANGPTL3 supposed to be expressed?
