Background of BRCA2

BRCA2 is a human tumor suppressor gene(specifically, a caretaker gene), found in all humans. This gene is mapped to 13q13.1. BRCA2 is essential for efficient homology-directed repair, it regulates both the intracellular localization and DNA-binding ability of RAD51. What's more, the complete BRC repeat domain of BRCA2 shows robust RAD51 binding and it is a critical cofactor for RAD51-mediated homologous recombination. BRCA2 is also a key mediator of homologous recombination and provides a molecular basis for understanding how this DNA repair process is disrupted by BRCA2 mutations. BRCA2-deficient cells, as a result of their deficiency in homologous recombination, are acutely sensitive to PARP inhibitors.
