Background of EIF2S2
Eukaryotic translation initiation factor 2 subunit 2, also called EIF2S2 is a protein that in humans is encoded by the EIF2S2 gene. This gene belongs to the EIF-2-beta family. EIF2S2 is mapped to 20q11.2. Eukaryotic translation initiation factor 2 (EIF-2) functions in the early steps of protein synthesis by forming a ternary complex with GTP and initiator tRNA and binding to a 40S ribosomal subunit. EIF-2 is composed of three subunits, alpha, beta, and gamma, with the protein encoded by this gene representing the beta subunit. The beta subunit catalyzes the exchange of GDP for GTP, which recycles the EIF-2 complex for another round of initiation.

Western blot analysis of EIF2S2 using anti-EIF2S2 antibody (PA2157).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: human HepG2 whole cell lysates,
Lane 2: human 293T whole cell lysates,
Lane 3: human Hela whole cell lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-EIF2S2 antigen affinity purified polyclonal antibody (Catalog # PA2157) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for EIF2S2 at approximately 38 kDa. The expected band size for EIF2S2 is at 38 kDa.

We are currently using anti-EIF2S2 antibody PA2157 for rat tissue, and we are satisfied with the WB results. The species of reactivity given in the datasheet says human, mouse, rat. Is it possible that the antibody can work on dog tissues as well?

The anti-EIF2S2 antibody (PA2157) has not been validated for cross reactivity specifically with dog tissues, but there is a good chance of cross reactivity.
