Background of IL6
Interleukin-6 (IL-6) is a protein that in humans is encoded by the IL6 gene. IL-6 is an interleukin that acts as both a pro-inflammatory and anti-inflammatory cytokine. It is secreted by T cells and macrophages to stimulate immune response to trauma, especially burns or other tissue damage leading to inflammation. IL-6 is one of the most important mediators of fever and of the acute phase response. IL-6 is also essential for hybridoma growth and is found in many supplemental cloning media such as briclone. Bowcock et al. (1988) assigned the IL6 gene to chromosome 7p21. By in situ hybridization and Southern blot analysis of mouse-human hybrid cell lines, Sutherland et al. (1988) mapped the IL-6 gene to chromosome 7p15.

According to literature, left coronary artery does express IL6. Generally IL6 expresses in secreted. Regarding which tissues have IL6 expression, here are a few articles citing expression in various tissues:

Question
Hi Support: I purchased the antibody against IL-6 with the SKU M00102-2 and used it as primary antibody in Western Blot. However, my results obtained 8 colored bands on the membrane (24 to 150kD), in which only one of them corresponds to the right molecular weight (about 24 kD). I want to figure out whether it is non-specific binding or not. Have you encountered the same profile before? Do you know which site of IL-6 is targeted by this antibody SKU M00102-2?

Answer
The mature protein of IL-6 can be highly N-glycosylated so the actual band is much higher than the expected. The sizes of the bands depend on the complexity of the polysaccharide chain. May I know what protocol you used in Western blot, to see if any alterations/optimizations are necessary?

We are currently using anti-human IL6 antibody (monoclonal, 7D12) M00102-2 for human tissue, and we are happy with the ELISA results. The species of reactivity given in the datasheet says human. Is it true that the antibody can work on canine tissues as well?

From what I have seen in literature left coronary artery does express IL6. From what I have seen in Uniprot.org, IL6 is expressed in left coronary artery, fibroblast, lung, among other tissues.
