Flow Cytometry analysis of HepG2 cells using anti-ACAA2 antibody (PB10022).
Overlay histogram showing HepG2 cells stained with PB10022 (Blue line). To facilitate intracellular staining, cells were fixed with 4% paraformaldehyde and permeabilized with permeabilization buffer. The cells were blocked with 10% normal goat serum. And then incubated with rabbit anti-ACAA2 Antibody (PB10022,1μg/1x106 cells) for 30 min at 20°C. DyLight®488 conjugated goat anti-rabbit IgG (BA1127, 5-10μg/1x106 cells) was used as secondary antibody for 30 minutes at 20°C. Isotype control antibody (Green line) was rabbit IgG (1μg/1x106) used under the same conditions. Unlabelled sample without incubation with primary antibody and secondary antibody (Red line) was used as a blank control.

Background of ACAA2
3-Ketoacyl-CoA thiolase, mitochondrial, also known as acetyl-Coenzyme A acyltransferase 2, is an acetyl-CoA C-acyltransferase enzyme that in humans is encoded by the ACAA2 gene. The ACAA2 gene encodes a 41.9 kDa protein that is composed of 397 amino acids and contains 88 observed peptides. The encoded protein catalyzes the last step of themitochondrial fatty acid beta oxidation spiral. Unlike most mitochondrial matrix proteins, it contains a non-cleavable amino-terminal targeting signal. Additionally, ACAA2 has been shown to be a functional BNIP3 binding partner, which provides a possible link between fatty acid metabolism and cell apoptosis.
