Background of CDK1
p34 cdc2 is a serine-threonine protein kinase of 34,000 daltons that complexes with cyclin to form maturation promoting factor (MPF). The inactive form of the protein is phosphorylated at threonine (T) and tyrosine (Y) residues. In humans the phosphorylation appears to be performed by p60src. The active form of the protein is dephosphorylated and it functions by phosphorylating a number of proteins. The phosphorylation activity is coupled to the entry into the M-phase of the cell. p34 cdc2 protein must be associated with a normal cyclin protein for the M-phase to be completed normally. Association with deletion mutants of cyclin halts the M-phase before it is completed.

Anti-p34 cdc2 antibody has been tested in ELISA and western blot and is suitable for immunoblotting, immunohistochemistry immunoprecipitation (as active kinase), and immunoblotting. HeLa cell lysate or human colon carcinoma is suggested as a positive control for immunoblotting. LEP fibroblast cell lysate is suggested as a negative control. This product is suitable for the detection by immunoblot of human, rat and mouse cdc2. For immunohistochemistry use paraffin embedded tissue.

Boster's anti-cdc2 Cyclin Dependent Kinase was used to detect human p34cdc2 by western blot in untreated (Control) and drug treated lysates of MCF-7 cells. Lane 1-4 represents 3.1µM, 6.2µM, 12.5µM and 25.0µM genistein treatment of cells before lysates were prepared. Detection occurs using a 1:1,000 dilution. Although this antiserum detects non-specific bands at higher MW, a clear induction of signal is observed as the concentration of drug is increased. Can you help my question with product A30951, anti-Cdc2 P34 antibody. I was wondering if it would be possible to conjugate this antibody with biotin. I would need it to be without BSA or sodium azide. I am planning on using a buffer exchange of sodium azide with PBS only. Would there be problems for me to conjugate the antibody and store it in -20 degrees in small aliquots?

We do not advise storing this antibody with PBS buffer only in -20 degrees. If you want to store it in -20 degrees it is best to add some cryoprotectant like glycerol. If you want carrier free A30951 anti-Cdc2 P34 antibody, we can provide it to you in a special formula with trehalose and/or glycerol. These molecules will not interfere with conjugation chemistry and provide a good level of protection for the antibody from degradation. Please be sure to specify this in your purchase order.

It shows on the product datasheet, A30951 anti-Cdc2 P34 antibody as been tested on WB. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.

We are currently using anti-Cdc2 P34 antibody A30951 for human tissue, and we are satisfied with the IP results. The species of reactivity given in the datasheet says human, mouse, rat. Is it likely that the antibody can work on primate tissues as well?
