Western blot analysis of NR2F1 using anti-NR2F1 antibody (M03833).
Electrophoresis was performed on a 10% SDS-PAGE gel at 80V (Stacking gel) / 120V (Resolving gel) for 2 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: human 293T whole cell lysates,
Lane 2: human PC-3 whole cell lysates,
Lane 3: human SH-SY5Y whole cell lysates,
Lane 4: human Hela whole cell lysates,
Lane 5: rat brain tissue lysates,
Lane 6: rat C6 whole cell lysates,
Lane 7: mouse brain tissue lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-NR2F1 antigen affinity purified monoclonal antibody (M03833) at 1:500 overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an ECL Plus Western Blotting Substrate (Catalog # AR1196-200) with Tanon 5200 system. A specific band was detected for NR2F1 at approximately 46 kDa. The expected band size for NR2F1 is at 46 kDa.

The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 1:50 rabbit anti-NR2F1 Antibody (M03833) overnight at 4°C. Peroxidase Conjugated Goat Anti-rabbit IgG was used as secondary antibody and incubated for 30 minutes at 37°C. The tissue section was developed using HRP Conjugated Rabbit IgG Super Vision Assay Kit (Catalog # SV0002) with DAB as the chromogen.

Does anti-COUP TF1 Monoclonal antibody M03833 work on bovine WB with lung uterus?

Our lab technicians have not validated anti-COUP TF1 Monoclonal antibody M03833 on bovine. You can run a BLAST between bovine and the immunogen sequence of anti-COUP TF1 Monoclonal antibody M03833 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated bovine samples, this use of the antibody is not covered by our guarantee. However we have an innovator award program that if you test this antibody and show it works in bovine lung uterus in WB, you can get your next antibody for free.

Question
We are currently using anti-COUP TF1 Monoclonal antibody M03833 for rat tissue, and we are satisfied with the IF results. The species of reactivity given in the datasheet says human, mouse, rat. Is it likely that the antibody can work on primate tissues as well?

The anti-COUP TF1 Monoclonal antibody (M03833) has not been tested for cross reactivity specifically with primate tissues, though there is a good chance of cross reactivity.
