This affinity purified antibody was prepared from whole rabbit serum produced by repeated immunizations with a synthetic peptide corresponding to an internal region near aa 150-175 of Human DNMT3L (DNA (cytosine-5-)-methyltransferase 3-like). DNMT3L is a nuclear protein with similarity to DNA methyltransferases.

Background of DNMT3L
DNMT 3L is a nuclear protein that has similarity to DNA methyltransferases, involved in de novo methylation of CpG islands. CpG methylation is an epigenetic modification that is important for embryonic development, imprinting, and X-chromosome inactivation. Studies in mice have demonstrated that DNA methylation is required for mammalian development. This gene encodes a nuclear protein with similarity to DNA methyltransferases. This protein is not thought to function as a DNA methyltransferase as it does not contain the amino acid residues necessary for methyltransferase activity. However, this protein does stimulate de novo methylation by DNA cytosine methyltransferase 3 alpha and it is thought to be required for the establishment of maternal genomic imprints. This protein also mediates transcriptional repression through interaction with histone deacetylase 1.

Anti-DNMT3L antibody has been tested for use in ELISA against the immunizing peptide. This antibody has been tested in immunohistochemistry and western blot. This antibody works for western blotting only following immunoprecipitation. The expected molecular weight of human DNMT3L is 43.6 kDa. Reactivity in other immunoassays is unknown.

Affinity Purified Rabbit anti-DNMT3L was used at a 1:1,500 dilution to detect human DNMT3L by western blot after immunoprecipitation using the same antibody. Transfection of U2OS cells (10cm dish) was accomplished using 3 µg of GAL4-DNMT3L. Protein extraction using IPH at 150mM. For IP and WB conditions see Fuks et al. (2000). For western blotting the antibody was used at 0.4µg/ml in TBS milk 2%, BSA 0.5%. Detection occurred using ECL.

The conserved N-terminal cysteine-rich region, the PHD-like motif, characteristic of the Dnmt3 family is shown. The C-terminal domain is the catalytic methyltransferase domain present in Dnmt3a and Dnmt3b whereas the key catalytic motifs are absent in Dnmt3L. Numbers indicate amino acid residues.

It shows on the product datasheet, A04214 anti-DNMT3L antibody as been tested on WB. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.

Our lab technicians have not validated anti-DNMT3L antibody A04214 on zebrafish. You can run a BLAST between zebrafish and the immunogen sequence of anti-DNMT3L antibody A04214 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated zebrafish samples, this use of the antibody is not covered by our guarantee. However we have an innovator award program that if you test this antibody and show it works in zebrafish placenta in WB, you can get your next antibody for free.

I have a question about product A04214, anti-DNMT3L antibody. I was wondering if it would be possible to conjugate this antibody with biotin. I would need it to be without BSA or sodium azide. I am planning on using a buffer exchange of sodium azide with PBS only. Would there be problems for me to conjugate the antibody and store it in -20 degrees in small aliquots?

We suggest not storing this antibody with PBS buffer only in -20 degrees. If you want to store it in -20 degrees it is best to add some cryoprotectant like glycerol. If you want carrier free A04214 anti-DNMT3L antibody, we can provide it to you in a special formula with trehalose and/or glycerol. These molecules will not interfere with conjugation chemistry and provide a good level of protection for the antibody from degradation. Please be sure to specify this in your purchase order.
