Background of Fcgrt
IgG receptor FcRn large subunit p51 is a protein that in humans is encoded by the FCGRT gene. This gene encodes a receptor that binds the Fc region of monomeric immunoglobulin G. The encoded protein transfers immunoglobulin G antibodies from mother to fetus across the placenta. This protein also binds immunoglobulin G to protect the antibody from degradation. In addition, it is postulated that long-range cis inactivation of the FCGRT gene is responsible for hypercatabolism of IgG in myotonic dystrophy. The FCGRT gene is closely situated to the DMPK gene, which is mutant in DM.
Antibody Validation
Boster validates all antibodies on WB, IHC, ICC, Immunofluorescence, and ELISA with known positive control and negative samples to ensure specificity and high affinity, including thorough antibody incubations.

Western blot analysis of FCGRT using anti-FCGRT antibody (PB9867).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: mouse liver tissue lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-FCGRT antigen affinity purified polyclonal antibody (Catalog # PB9867) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for FCGRT at approximately 50 kDa. The expected band size for FCGRT is at 40 kDa.

IHC analysis of FCGRT using anti-FCGRT antibody (PB9867).
FCGRT was detected in a paraffin-embedded section of mouse brain tissue. Heat mediated antigen retrieval was performed in EDTA buffer (pH 8.0, epitope retrieval solution). The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 1 μg/ml rabbit anti-FCGRT Antibody (PB9867) overnight at 4°C. Biotinylated goat anti-rabbit IgG was used as secondary antibody and incubated for 30 minutes at 37°C. The tissue section was developed using Strepavidin-Biotin-Complex (SABC) (Catalog # SA1022) with DAB as the chromogen.

7 Customer Q&As for Anti-FCGRT Antibody Picoband®
Question
Would PB9867 anti-FCGRT antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?
Verified Customer
Verified customer
Asked: 2020-04-21
Answer
As indicated on the product datasheet, PB9867 anti-FCGRT antibody as been validated on WB. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.

Question
Will anti-FCGRT antibody PB9867 work on feline WB with placenta?
T. Mitchell
Verified customer
Asked: 2017-04-10
Answer
Our lab technicians have not validated anti-FCGRT antibody PB9867 on feline. You can run a BLAST between feline and the immunogen sequence of anti-FCGRT antibody PB9867 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated feline samples, this use of the antibody is not covered by our guarantee. However we have an innovator award program that if you test this antibody and show it works in feline placenta in WB, you can get your next antibody for free.
