Background of LHCGR
The luteinizing hormone/choriogonadotropin receptor is a member of a subfamily of G protein-coupled receptors (GPCR) characterized by the presence of a large N-terminal extracellular domain containing several leucine-rich repeats (LRR). This glycoprotein hormone receptor family has been named the LRR-containing GPCR (LGR) family. The LHCGR gene contains 11 exons and spans approximately 80 kb and it is mapped to chromosome 2p21. In the ovary, theca, stromal, late-stage (luteinizing) granulosa, and luteal cells contain LHCGR. In the testes, Leydig cells contain LHCRG.
Antibody Validation
Boster validates all antibodies on WB, IHC, ICC, Immunofluorescence, and ELISA with known positive control and negative samples to ensure specificity and high affinity, including thorough antibody incubations.

IHC analysis of LHCGR using anti-LHCGR antibody (PA1552).
LHCGR was detected in a paraffin-embedded section of human ovarian cancer tissue. Heat mediated antigen retrieval was performed in EDTA buffer (pH 8.0, epitope retrieval solution). The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 2 μg/ml rabbit anti-LHCGR Antibody (PA1552) overnight at 4°C. Peroxidase Conjugated Goat Anti-rabbit IgG was used as secondary antibody and incubated for 30 minutes at 37°C. The tissue section was developed using HRP Conjugated Rabbit IgG Super Vision Assay Kit (Catalog # SV0002) with DAB as the chromogen.

Distribution of aldosterone synthase (CYP11B2) and LHCGR in the patient’s adrenal. (A) , Distribution of CYP11B2 immunoreactivity in the tumor region (T) of the adrenal tissue. (B-E) , Distribution of CYP11B2 (B, D) and LHCGR (C, E) immunoreactivities in consecutive sections of the APA tissue at low (B, C) and high (D, E) magnifications. Similar distribution of CYP11B2 and LHCGR immunoreactivities were observed in some areas (arrows) in (B, C) . PT indicates peritumoral tissue; and V, vein.

Asked: 2019-10-21
Answer
Our technicians provide these suggestions to reduce the background: 1.Block the membrane 2 hours at room temperature with 5% no-fat milk in TBS-T; 2.Dilute second antibody with 5% no-fat milk in TBS-T; and 3.Extend the wash time to 10 minutes each time.

Question
We are currently using anti-hCG receptor/LHCGR antibody PA1552 for rat tissue, and we are happy with the IHC results. The species of reactivity given in the datasheet says human, rat. Is it possible that the antibody can work on bovine tissues as well?
Verified Customer

Asked: 2019-05-20
Answer
The anti-hCG receptor/LHCGR antibody (PA1552) has not been tested for cross reactivity specifically with bovine tissues, though there is a good chance of cross reactivity. We have an innovator award program that if you test this antibody and show it works in bovine you can get your next antibody for free. Please contact me if I can help you with anything.
