Background of HMGN1

The specificity of mAb IPO-38 was demonstrated in different human and murine lymphoid and non-lymphoid cell lines. It does not react with mononuclear cells and granulocytes of healthy donors but does react with nuclei of blood cells of patients with different forms of leukemia and with cells from lymph nodes of patients with Hodgkin's disease and non-hodgkin's lymphoma. Also a positive reaction in nuclei of breast and colorectal tumor cells was demonstrated. IPO-38 may be a good marker of cellular proliferation during monitoring of tumor progression.

Antibody Validation

Boster validates all antibodies on WB, IHC, ICC, Immunofluorescence, and ELISA with known positive control and negative samples to ensure specificity and high affinity, including thorough antibody incubations.

Assay Dilutions Recommendation

The recommendations below provide a starting point for assay optimization. The actual working concentration varies and should be decided by the user.

ELISA: 1:10,000 - 1:50,000
IHC: 10 µg/mL
WB: 1:500- 1:2,000
Anti-IPO-38 has been tested by immunohistochemistry (frozen and formalin/paraffin) and western blot analysis on the Raji cell line and reacts with a 14 -16 kDa protein. This antibody is suitable for ELISA and immunoprecipitation.  The antibody is reported to recognize a nuclear antigen that is present in the cytoplasm and nuclei of proliferating cells (paraformaldehyde fixed, Triton X-100 permeabilized).  An increase of 400% is observed in cells in mitosis (determined by analysis of K562 cells synchronized with colcemid).  IPO-38 does not block the binding of the Ki-67 antibody.   Tonsillar tissue is typically used as a positive control.

Question

I have a question about product M03051, anti-IPO-38 Monoclonal antibody. I was wondering if it would be possible to conjugate this antibody with biotin. I would need it to be without BSA or sodium azide. I am planning on using a buffer exchange of sodium azide with PBS only. Would there be problems for me to conjugate the antibody and store it in -20 degrees in small aliquots?

K. Dhar

Asked: 2015-03-30

Answer

We suggest not storing this antibody with PBS buffer only in -20 degrees. If you want to store it in -20 degrees it is best to add some cryoprotectant like glycerol. If you want carrier free M03051 anti-IPO-38 Monoclonal antibody, we can provide it to you in a special formula with trehalose and/or glycerol. These molecules will not interfere with conjugation chemistry and provide a good level of protection for the antibody from degradation. Please be sure to specify this in your purchase order.

Asked: 2013-03-20

Answer

The anti-IPO-38 Monoclonal antibody (M03051) has not been validated for cross reactivity specifically with horse tissues, but there is a good chance of cross reactivity. We have an innovator award program that if you test this antibody and show it works in horse you can get your next antibody for free. Please contact me if I can help you with anything.
