Background of AP2M1
AP-2 complex subunit mu is a protein that in humans is encoded by the AP2M1 gene. This gene encodes a subunit of the heterotetrameric coat assembly protein complex 2 (AP2), which belongs to the adaptor complexes medium subunits family. The encoded protein is required for the activity of a vacuolar ATPase, which is responsible for proton pumping occurring in the acidification of endosomes and lysosomes. The encoded protein may also play an important role in regulating the intracellular trafficking and function of CTLA-4 protein. Three transcript variants encoding different isoforms have been found for this gene.
Boster validates all antibodies on WB, IHC, ICC, Immunofluorescence and ELISA with known positive and negative samples to ensure specificity and high affinity.

Figure 1. Western blot analysis of AP2M1 using anti-AP2M1 antibody (A06179).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
Lane 1: human U2OS whole cell lysates,
Lane 2: human MCF-7 whole cell lysates,
Lane 3: rat kidney tissue lysates,
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-AP2M1 antigen affinity purified polyclonal antibody (Catalog # A06179) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for AP2M1 at approximately 50KD. The expected band size for AP2M1 is at 50KD.
