Background of Mre11
Mre11 is a component of the MRN complex (Mre11/Rad50/Nbs1), which plays a central role in double-strand break (DSB) repair, DNA recombination, maintenance of telomere integrity and meiosis. The complex possesses single-strand endonuclease activity and double-strand-specific 3'-5' exonuclease activity, which are provided by MRE11A. RAD50 may be required to bind DNA ends and hold them close. This could facilitate searches for short or long regions of sequence homology in the recombining DNA templates, and may also stimulate the activity of DNA ligases and/or restrict the nuclease activity of MRE11A to prevent nucleolytic degradation past a given point. The complex may also be required for DNA damage signaling via activation of the ATM kinase. In telomeres, the MRN complex may modulate t-loop formulation. Anti-Mre11 Antibody is useful for researcher interested in DNA Damage and Repair and Nuclear Signaling Research.

Assay Dilutions Recommendation
The recommendations below provide a starting point for assay optimization. The actual working concentration varies and should be decided by the user.
ELISA: 1:10,000 - 1:50,000
WB: 1:500 - 1:2,000
This affinity purified antibody has been tested for use in ELISA and western blot. Specific conditions for reactivity should be optimized by the end user. Expect a band approximately 80 kDa in size corresponding to Mre11 by western blotting in the appropriate cell lysate or extract.
Validation Images & Assay Conditions

Western blot using Boster's Affinity Purified anti-Mre11 antibody shows detection of a band ~80 kDa corresponding to mouse Mre11 (arrowhead). Lanes 1-4 contain 0.5 ug, 0.3 ug, 0.1 ug and 0.05 ug of purified mouse Mre11 protein, respectively. After 4-20% SDS-PAGE and transfer onto nitrocellulose, the membrane was blocked and then probed with the primary antibody diluted to 1:1,000 overnight at 4°C. The membrane was then washed and reacted with a 1:10,000 dilution of IRDye800 conjugated Gt-a-Rabbit IgG [H&L] MX (611-132-122) for 45 min at room temperature. IRDye800 fluorescence image was captured using the Odyssey® Infrared Imaging System developed by LI-COR. IRDye is a trademark of LI-COR, Inc. Other detection systems will yield similar results.

6 Customer Q&As for Anti-Mre11 Antibody
Question
Will A00731 anti-Mre11 antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?
Verified Customer

Asked: 2020-04-09
Answer
You can see on the product datasheet, A00731 anti-Mre11 antibody as been validated on WB. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.

Question
Can you help my question with product A00731, anti-Mre11 antibody. I was wondering if it would be possible to conjugate this antibody with biotin. I would need it to be without BSA or sodium azide. I am planning on using a buffer exchange of sodium azide with PBS only. Would there be problems for me to conjugate the antibody and store it in -20 degrees in small aliquots?
Verified Customer

Asked: 2019-10-11
Answer
We do not advise storing this antibody with PBS buffer only in -20 degrees. If you want to store it in -20 degrees it is best to add some cryoprotectant like glycerol. If you want carrier free A00731 anti-Mre11 antibody, we can provide it to you in a special formula with trehalose and/or glycerol. These molecules will not interfere with conjugation chemistry and provide a good level of protection for the antibody from degradation. Please be sure to specify this in your purchase order.
