Background of CASP10
Caspase-10 is an enzyme that, in humans, is encoded by the CASP10 gene. The Caspase 10 gene contains 11 exons and spans about 48 kb. This gene is mapped to 2q33.1. It is transcribed in the centromere-to-telomere direction.This gene encodes a protein that is a member of the cysteine-aspartic acid protease (caspase) family. Sequential activation of caspases plays a central role in the execution-phase of cell apoptosis. Caspases exist as inactive proenzymes that undergo proteolytic processing at conserved aspartic residues to produce two subunits, large and small, that dimerize to form the active enzyme. This protein cleaves and activates caspases 3 and 7, and the protein itself is processed by caspase 8. Mutations in this gene are associated with apoptosis defects seen in type II autoimmune lymphoproliferative syndrome. Three alternatively spliced transcript variants encoding different isoforms have been described for this gene.

We are currently using anti-Caspase-10/CASP10 antibody PA1513 for human tissue, and we are happy with the WB results. The species of reactivity given in the datasheet says human. Is it possible that the antibody can work on horse tissues as well?

The anti-Caspase-10/CASP10 antibody (PA1513) has not been validated for cross reactivity specifically with horse tissues, but there is a good chance of cross reactivity. Question
I got a high background from the bands on my western blot gel. Here are the parameters that I used: - Protein loading 70 µg - SDS gel 10% - Electrophoresis 60V 60 min & 120V 95 min - Blocking 5% milk RT 30 min - Primary antibody 1:2000 dilution & 4°C overnight incubation - Secondary antibody α-Rabbit –HRP RT incubation for 1 hour. Please advise how we can lower the background.

Answer
Try to repeat your western blot with the following changes: - Protein loading 50 µg - Blocking 5% non-fat dry milk & incubate at 4°C for 2 hours with slow shaking - Primary antibody 1:4000 dilution. Then the background of your gel should be reduced.
