Background of Il7
IL-7, Interleukin 7, is a protein that in humans is encoded by the IL-7 gene. IL-7 a hematopoietic growth factor secreted by stromal cells in the red marrow and thymus. It is also produced by keratinocytes, dendritic cells, hepatocytes, neurons, and epithelial cells but is not produced by lymphocytes. By the combination of approaches, IL-7 is located on 8q12-q13. IL-7 is critical for early T-cell development and homeostasis of naive and memory CD8-positive T cells. It signals through the IL-7 receptor complex, which consists of the IL7R-alpha chain and the common IL2R-gamma chain is critical for early T-cell development and homeostasis of naive and memory CD8 -positive T cells. It signals through the IL-7 receptor complex, which consists of the IL7R-alpha chain and the common IL2R-gamma chain.

Western blot analysis of IL7 using anti-IL7 antibody (PA1467).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
Lane 1: human PC-3 whole cell lysates.
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-IL7 antigen affinity purified polyclonal antibody (Catalog # PA1467) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for IL7 at approximately 20KD. The expected band size for IL7 is at 20KD.

My boss were satisfied with the WB result of your anti-IL7 antibody. However we have been able to see positive staining in pancreas secreted. using this antibody. Is that expected? Could you tell me where is IL7 supposed to be expressed?

From what I have seen in literature, pancreas does express IL7. Generally IL7 expresses in secreted. Regarding which tissues have IL7 expression, here are a few articles citing expression in various tissues:

We are currently using anti-IL7 antibody PA1467 for mouse tissue, and we are content with the IHC-P results. The species of reactivity given in the datasheet says human, mouse. Is it true that the antibody can work on zebrafish tissues as well?
