Western blot analysis of IL7 using anti-IL7 antibody (PB9521).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours.
Lane 1: K562 Whole Cell Lysate at 40ug.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-IL7 antigen affinity purified polyclonal antibody (Catalog # PB9521) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for IL7 at approximately 75 kDa. The expected band size for IL7 is at 20 kDa.

Background of IL7
IL-7, Interleukin 7, is a protein that in humans is encoded by the IL-7 gene. IL-7 a hematopoietic growth factor secreted by stromal cells in the red marrow and thymus. It is also produced by keratinocytes, dendritic cells, hepatocytes, neurons, and epithelial cells but is not produced by lymphocytes. By the combination of approaches, IL-7 is located on 8q12-q13. IL-7 is critical for early T-cell development and homeostasis of naive and memory CD8-positive T cells. It signals through the IL-7 receptor complex, which consists of the IL7R-alpha chain and the common IL2R-gamma chain is critical for early T-cell development and homeostasis of naive and memory CD8 -positive T cells. It signals through the IL-7 receptor complex, which consists of the IL7R-alpha chain and the common IL2R-gamma chain.
