Facts about AP-2 complex subunit alpha-1.
Adaptor protein complexes are vesicle coat components and seem to be involved in cargo selection and vesicle formation. AP-2 is involved in clathrin-dependent endocytosis where cargo proteins are incorporated into vesicles surrounded by clathrin (clathrin- coated vesicles, CCVs) that are destined for fusion with the early endosome.

The AP2A1 gene is a protein that binds to antigens and AP2A1 is a good candidate for such a specificity. It is not always desirable for a protein to have a high affinity towards antigens. This makes the separation of antigen and antibody more difficult. Therefore, high-affinity primary antibodies using the AP2A1 marker are highly useful in immunohistochemistry.
AP2A1, a common protein found on human cells' surfaces, is a common one. Many vesicles contain multiple epitopes which allow high-affinity primaries to recognize them. Purification of anti-IFN-g scFv was performed by Ni2+ affinity chromatography. These steps were similar to protein purification. The purified scFv products were tested for activity using ELISA. IFN-g antigen was coated onto 96-well plates overnight at 4degC. Antibodies were then added to the reaction wells, followed by addition of anti-MBP tag antibodies. Finally, HRP-conjugated goat IgG antibody against anti-mouse was used to detect the purified SCFv products.

The resulting plasma-derived complexes were sorted by antibodies against Argonaute 2 or PCSA. These double positive events were further quantified through flow cytometry. To study the effect of IFNg on the immune system, immunoassays using IFNg detection have been employed. This antibody is able to detect the antibody to IFN-g without the need to stimulate the immune systems.
