Ficolin-1 (FCN1)

Extracellular lectin functioning as a pattern- recognition receptor in innate immunity. Binds the sugar moieties of pathogen-associated molecular patterns (PAMPs) displayed on microbes and activates the lectin pathway of the complement system.

May also activate monocytes via a G protein-coupled receptor, FFAR2, inducing the secretion of interleukin-8/IL-8 (PubMed:21037097). Binds preferentially to 9-O-acetylated 2-6- linked sialic acid derivatives and to various glycans containing sialic acid participated in a 2-3 linkage.

Function

The FCN1N is associated with IgG1 by IgG-3D or Ch2p-3. This interaction has important implications in cellular and tissue immunology. In vitro, FCN1-N directly bound to the IgG1-3d domain. In a study involving three peptides, FCN1N only bound to IgG1. However, FCN1-IgG1 inhibition varied with dissection sites. Future studies will reveal the precise domains involved.

In patients with KD, serum FCN1 level decreased dramatically after IVIG therapy. These results indicate that IVIG therapy can target the FCN1 gene in a crucial way. Further studies will need to identify the precise role of this marker in determining the severity of KD. FCN1 expression is also associated with a host of other biomarker gene genes, including IL-10 (CCL18), IL-26 (CCL18), FABP4 and CCL2 (2 CCL18).

BALF's FCN1 positive macrophages are similar to pathogenic ST CD48highS100A12 clusters, which contributes to the severity COVID-19. FCN1+ macrophages are also similar to those that resolve TREM2pos macrophages. Furthermore, FCN1-positive macrophages have a similar transcriptional profile to resolving TREM2pos macrophages, which may indicate that they share homeostatic mechanisms.

FCN1-positive monocytes undergo a transition from monocyte derived macrophages into tissue-resident macrophages. After the transition, they undergo a series IL-1B, CCL2-positive, monocyte-derived macrophage formation events. FCN1-positive monocytes eventually will become alveolar microphages. This lineage will eventually mature to T h2 and Th27 macrophages.

Applications

There are several polymorphisms within the FCN1 gene. One such polymorphism is found at nucleic Acid position -1981 within the gDNA sequencing. This polymorphism also includes complement G > A. It is identified in a SNP databank as rs2989727. Other variants have also been identified as: rs2989690 e rs2989798, respectively.

The FCN1 genome is associated with IgG via the IgG-3–d and IgGp-3 domains. However, the FCN1 gene has many applications, including stratification of risk for rheumatoid arthritis. It is also known how many copies are associated with rheumatoid. The FCN1 gene polymorphisms, which are found in the sequences of RNA and DNA, could also be used as a way to identify rheumatoid.
