Background of Cofilin

Cofilin, a member of the ADF/cofilin family, is actually a protein with 70% sequence homology to ADF, making it part of the ADF/cofilin family of small ADP-binding proteins. It is a ubiquitous actin-binding factor required for the reorganization of actin filaments. The protein is known to sever actin filaments by creating more positive ends on filament fragments, it can cause depolymerization at the minus end of filaments, thereby preventing their reassembly. As a long-lasting in vivo effect, cofilin recycles older ADP-F-actin, helping cell to maintain ATP-G-actin pool for sustained motility. pH, phosphorylation and phosphoinositides regulate cofilin’s binding and associating activity with actin.

Antibody Validation

Boster validates all antibodies on WB, IHC, ICC, Immunofluorescence, and ELISA with known positive control and negative samples to ensure specificity and high affinity, including thorough antibody incubations.

5 Customer Q&As for Anti-Cofilin/CFL1 Antibody Picoband™

Question

We bought anti-Cofilin/CFL1 antibody for WB on pre-b cell in the past. I am using rat, and We intend to use the antibody for IHC-P next. I was wanting to use examining pre-b cell as well as b-cell lymphoma in our next experiment. Could you please give me some suggestion on which antibody would work the best for IHC-P?

Verified Customer

Answer

Based on literature lung does express CFL1. Based on Uniprot.org, CFL1 is expressed in cerebellum, placenta, pre-b cell, testis, lung, ovary, placenta uterus, platelet, b-cell lymphoma, brain, cajal-retzius cell fetal brain cortex, lymphoblast, cervix carcinoma, leukemic t-cell, cervix carcinoma erythroleukemia, liver, among other tissues. Regarding which tissues have CFL1 expression, here are a few articles citing expression in various tissues:

Question

We are currently using anti-Cofilin/CFL1 antibody PB9033 for mouse tissue, and we are happy with the Flow Cytometry results. The species of reactivity given in the datasheet says human, monkey, mouse, rat. Is it possible that the antibody can work on pig tissues as well?

Verified Customer

Asked: 2018-01-11

Answer

The anti-Cofilin/CFL1 antibody (PB9033) has not been tested for cross reactivity specifically with pig tissues, though there is a good chance of cross reactivity. We have an innovator award program that if you test this antibody and show it works in pig you can get your next antibody for free. Please contact me if I can help you with anything.
