Background of HOXA3

HOXA3 (HOMEOBOX A3) is a protein that in humans is encoded by the HOXA3 gene, which is also part of the A cluster on chromosome 7. HOXA3 encodes a DNA-binding transcription factor which may regulate gene expression, morphogenesis, and differentiation. Its cytogenetic location is 7q15.2. In vertebrates, the genes encoding the class of transcription factors called homeobox genes are found in clusters named A, B, C, and D on four separate chromosomes. Expression of these proteins is spatially and temporally regulated during embryonic development. HOXA3 protein is able to complement Hoxd3 deficiency when expressed in the context of the Hoxd3 allele.

Would PA1602 anti-HOXA3 antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?

Answer

You can see on the product datasheet, PA1602 anti-HOXA3 antibody as been tested on WB. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.

Our lab technicians have not validated anti-HOXA3 antibody PA1602 on zebrafish. You can run a BLAST between zebrafish and the immunogen sequence of anti-HOXA3 antibody PA1602 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated zebrafish samples, this use of the antibody is not covered by our guarantee. However we have an innovator award program that if you test this antibody and show it works in zebrafish female gonad in WB, you can get your next antibody for free.
