Background of MRPL20

Mammalian mitochondrial ribosomal proteins are encoded by nuclear genes and help in protein synthesis within the mitochondrion. Mitochondrial ribosomes (mitoribosomes) consist of a small 28S subunit and a large 39S subunit. They have an estimated 75% protein to rRNA composition compared to prokaryotic ribosomes, where this ratio is reversed. Another difference between mammalian mitoribosomes and prokaryotic ribosomes is that the latter contain a 5S rRNA. Among different species, the proteins comprising the mitoribosome differ greatly in sequence, and sometimes in biochemical properties, which prevents easy recognition by sequence homology. This gene encodes a 39S subunit protein. A pseudogene corresponding to this gene is found on chromosome 21q.

We are currently using anti-MRPL20 antibody A14230 for mouse tissue, and we are content with the WB results. The species of reactivity given in the datasheet says human, mouse, rat. Is it possible that the antibody can work on primate tissues as well?

The anti-MRPL20 antibody (A14230) has not been tested for cross reactivity specifically with primate tissues, though there is a good chance of cross reactivity. Does A14230 anti-MRPL20 antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?

Answer

You can see on the product datasheet, A14230 anti-MRPL20 antibody as been validated on WB. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.
