Background of SIGLEC5

Mediator of RNA polymerase II transcription subunit 4, also known as mediator complex subunit 4 (MED4), a component of Mediator or vitamin D3 receptor-interacting protein complex 36 kDa component (DRIP36), is a protein that in humans is encoded by the MED4 gene. The Mediator complex interacts with DNA-binding gene-specific transcription factors to modulate transcription by RNA polymerase II. The MED4 gene was mapped to chromosome 13 based on sequence similarity between the MED4 sequence and a genomic contig.

Would A06510-1 anti-Siglec-5/14 antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?

Answer

You can see on the product datasheet, A06510-1 anti-Siglec-5/14 antibody as been tested on IHC. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.

We are currently using anti-Siglec-5/14 antibody A06510-1 for human tissue, and we are satisfied with the IHC results. The species of reactivity given in the datasheet says human. Is it likely that the antibody can work on bovine tissues as well?

The anti-Siglec-5/14 antibody (A06510-1) has not been tested for cross reactivity specifically with bovine tissues, though there is a good chance of cross reactivity. Our lab technicians have not validated anti-Siglec-5/14 antibody A06510-1 on goat. You can run a BLAST between goat and the immunogen sequence of anti-Siglec-5/14 antibody A06510-1 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated goat samples, this use of the antibody is not covered by our guarantee.
