Background of TMEM199
TMEM199 encodes a protein homologous to the yeast V-ATPase assembly factor Vma12 and appears to be involved in Golgi homeostasis. The protein encoded by this gene has been observed to localize to the endoplasmic reticulum (ER)-Golgi intermediate compartment (ERGIC) and coat protein complex I (COPI) in some human cells. Defects in this gene are a cause of congenital disorder of glycosylation, type IIp. By genomic sequence analysis, the TMEM199 gene is mapped to chromosome 17q11.1.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
Lane 1: human Hela whole cell lysates,
Lane 2: human placenta tissue lysates,
Lane 3: human Caco-2 whole cell lysates,
Lane 4: human T-47D whole cell lysates,
Lane 5: human U-87MG whole cell lysates,
Lane 6: human K562 whole cell lysates,
Lane 7: human U2OS whole cell lysates,
Lane 8: human PC-3 whole cell lysates.
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-TMEM199 antigen affinity purified polyclonal antibody (Catalog # A14686-1) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for TMEM199 at approximately 23KD. The expected band size for TMEM199 is at 23KD.

We are currently using anti-TMEM199 antibody A14686-1 for human tissue, and we are happy with the ELISA results. The species of reactivity given in the datasheet says human, mouse, rat. Is it true that the antibody can work on bovine tissues as well?

The anti-TMEM199 antibody (A14686-1) has not been tested for cross reactivity specifically with bovine tissues, though there is a good chance of cross reactivity.
