Background of RBP4
Retinol binding protein 4, plasma, also known as RBP4, belongs to the lipocalin family and is the specific carrier for retinol (vitamin A alcohol) in the blood. It is protein that in humans is encoded by the RBP4 gene. RBP4 gene resides just centromeric of the cluster of CYP2C genes on 10q24. The mouse Rbp4 locus is closely linked and just proximal to the locus for phenobarbital-inducible cytochrome P450-2c (Cyp-2c) at the distal end of chromosome 19. It delivers retinol from the liver stores to the peripheral tissues. In plasma, the RBP-retinol complex interacts with transthyretin, which prevents its loss by filtration through the kidney glomeruli. A deficiency of vitamin A blocks secretion of the binding protein posttranslationally and results in defective delivery and supply to the epidermal cells. The standard used in this kit is recombinant protein, with E19-L201 aa sequence, the molecular weight is 22kda.

7 Customer Q&As for Human RBP4/Retinol Binding Protein 4 ELISA Kit PicoKine®
Question
Q: how are cell lysates prepared for use in Picokine® ELISA kits?
S. Maharaj
Verified customer
Asked: 2020-09-12
Answer
A: in those Picokine® ELISAs where cell or tissue lysate is a validated sample type, sample preparation instructions for lysate are included in the product insert. Components in lysate and lysis buffer can change immunoreactivity, so if lysate is not a validated sample type, care must be taken in sample preparation and validation.

Question
Q: for how much duration can samples (cell cultures, serum, and plasma) be stored and still be stable for analysis RBP4 using the EK0831 Human RBP4 Picokine® ELISA Kit?
A. Surve
Verified customer
Asked: 2020-07-31
Answer
A: Boster Bio does not evaluate sample stability. Variations in sample collection, processing, and storage may change the stabilityof samples. It is recommend to assay sample on priority basis collection when possible, or aliquot into single use volumes and store samples frozen. avoid repeat freeze-thaw cycles with the stored samples to avoid protein degradation.

Question
Q: Are Boster Bio recombinant proteins and antibodies sterile?
Verified Customer
Verified customer
Asked: 2019-09-28
Answer
A: although the vials are bottled using aseptic techniques, heat-treated vials, and sterile stock solutions, they are not considered or guaranteed to be sterile. If sterile material is required for an experiment, the material can be filtered through a 0.2 micron filter designed for use with biological fluids.

Question
Q: which procedure should I follow in order to thaw whole blood sample for RBP4 ELISA after freezing?
K. Williams
Verified customer
Asked: 2018-09-04
Answer
A: we do not recommend freezing and thaw whole blood. erythrocytes are fragile and, if frozen and thawed, will undergo hemolysis rendering the samples useless. To keep your blood samples to test RBP4 for a later time, you should let the blood clot in glass tubes and separate the serum to freeze for later analysis.

Question
Q: can I use citrate plasma as samples in Human RBP4 Picokine® ELISA Kit (Catalog # EK0831)?
Verified Customer
Verified customer
Asked: 2017-11-13
Answer
A: Chelating agents such as EDTA, Heparin and Citrate can irreversibly bind metal ions from the functional domain of RBP4 causing degradation of its protein structure. RBP4 may be denatured as a result and may compromise the assay's measurements. The chilating sites could also be too close to the epitopes a must for detection and block the antigen antibody reaction. We have tested the RBP4 ELISA, treating samples with various anticoagulants and decided that heparin or EDTA can be used for treatment of blood/plasma samples. Do not use other anticoagulents when collecting samples.

Question
Q: we need your suggestion regarding the dilution ratio of serum samples for detection of RBP4 in Human urine? I am trying to measure a few analytes and it requires 100ul of diluted samples for each well. We have limited sample quantitys so we like to dilute as much as possible.
K. Anand
Verified customer
Asked: 2016-03-27
Answer
A: having little idea about the physiological or pathological context of your samples we cannot recommend a dilution ratio without performing a pilot test with your samples. Here is how you can perform a pilot study on your own: perform a serial dilution of your samples on the RBP4 ELISA kit to make sure you have a linear ascending curve followed by a plateau, which signifies the samples saturating the detection limit of the kit. Then you can pick the dilution ratios from samples in the linear part of the curve as your experimental dilution ratio.
If you are interested in using our ELISA service, you can also send us your sample and we will take care of everything for you. You can check our service details here: bosterbio.com/services/assay-services/elisa-testing-service
