Background of SDC1

Syndecan 1, also known as SYND1 or CD138, is a protein which in humans is encoded by the SDC1 gene. The protein encoded by this gene is a transmembrane (type I) heparan sulfate proteoglycan and is a member of the syndecan proteoglycan family. This gene is mapped to 2p24.1. The syndecans mediate cell binding, cell signaling, cytoskeletal organization and syndecan receptors are required for internalization of the HIV-1 tat protein. The syndecan 1 protein functions as an integral membrane protein and participates in cell proliferation, cell migration and cell-matrix interactions via its receptor for extracellular matrix proteins. What’s more, Syndecan-1 is also a sponge for growth factors, with binding largely via heparan sulfate chains.

Question

I did not get any measurable Syndecan-1 signal in my cell culture supernatants on ELISA but I expected some baseline signal even if it was small. Meanwhile, the standards worked fine, and I also confirmed cell activation by flow cytometry with ICAM-1. What happened? Any troubleshooting suggestions?

Answer

Confirmed cell activation doesn't indicate the sample contains protein of interest. The protein may also be in small quantities beyond the ELISA detection range. To troubleshoot, we suggest that you try to increase the cell density and/or homogenize/lyse the cells in lysis buffer (instead of working with cell supernatant).

Q: What is the optimal O.D. value for CD138 ELISA kit? I used your CD138 ELISA on serum samples. For my positive control, I received an O.D. value of 0.826, while my negative control received a value of 0.136. I obtained both of these controls from the ELISA kit, where your kit's typical data shows O.D. values much higher than my positive control and your background is lower. My samples O.D. values are around 0.225 and the highest is only 0.357. can I consider these samples contain CD138 even though the O.D. values are not very high?

A: The absolute O.D. values may change according to incubation time. The more you incubate the higher the O.D. values are going to be. a point of focus should be is whether your sample O.D. values are statistically significantly higher than your blank values. considering your assay, you could extend your development time in the substrate incubation step to obtain higher O.D. values, as long as your negative controls' O.D. values are not increasing faster in relation to your positive controls. typically, a sample with O.D. value 2 standard deviations higher than your negative controls can be considered positive. We calculate the sensitivity of this ELISA kit by converting cutoff O.D. value, calculated as the average of 20 negative controls plus 2 standard deviations of the 20 negative controls, into a concentration. in other words, when we claim this CD138 ELISA kit to have sensitivity of 10pg/ml, that means the minimum amount of CD138 that can be declared/interpreted as positive by the above standard is 10pg/ml.

A: in those Picokine® ELISAs where cell or tissue lysate is a validated sample type, sample preparation instructions for lysate are included in the product insert. Components in lysate and lysis buffer can effect immunoreactivity, so if lysate is not a validated sample type, care must be taken in sample preparation and validation.

A: Boster Bio does not evaluate sample stability. Variations in sample collection, processing, and storage may alter the stabilityof samples. It is recommend to assay sample immediately after collection when possible, or aliquot into single use volumes and store samples frozen. limit repetitive freeze-thaw cycles with the stored samples in order to prevent protein degradation.

Q: we need your recommendation regarding the dilution ratio of serum samples for detection of CD138 in Human serum? I am trying to measure a multiple analytes and it requires 100ul of diluted samples for each well. We have low serum quantitys so we like to dilute as much as possible.

A: unable to know the physiological or pathological context of your samples we cannot recommend a dilution ratio without performing a pilot test with your samples. Here is how you can perform a pilot study on your own: perform a serial dilution of your samples on the CD138 ELISA kit to make sure you have a linear ascending curve followed by a plateau, which signifies the samples saturating the detection limit of the kit. Then you can pick the dilution ratios from samples in the linear part of the curve as your experimental dilution ratio.
