Background of TNF RI/TNFRSF1A

TNFsRI (also known as Solube tumor necrosis factor receptor 1) is a cytokine receptor that binds tumor necrosis factors (TNF). In contrast, the p60 (TNFRSF1A) and p80 (TNFRSF1B) TNFA receptors self-assemble through a distinct functional domain in the TNFR extracellular domain, termed the pre-ligand assembly domain (PLAD), in the absence of ligand. Deletion of the PLAD results in monomeric presentation of p60 or p80. Flow cytometric analysis showed that efficient TNFA binding depends on receptor self-assembly. They also found that other members of the TNF receptor superfamily, including the extracellular domains of TRAIL (TNFRSF10A), CD40, and FAS (TNFRSF6), all self-associate but do not interact with heterologous receptors. By Southern blot analysis of human/Chinese hamster somatic cell hybrid DNA, the TNFR1 gene was mapped to 12pter-cen. And by nonradioactive in situ hybridization that the type 1 receptor (the p55 TNF receptor) is encoded by a gene located on chromosome 12p13.2.

9 Customer Q&As for Human TNFsR I ELISA Kit PicoKine®

Question

I have some inquiry regarding your product Human TNFsR I ELISA Kit PicoKine. Do you have any records about its endotoxin level? What protocol used in order to determine the antibody neutralization activity (it's probably proliferation of PBMC but I need the protocol). Is it azide free? Please advise.

Verified Customer

Verified customer

Asked: 2020-04-06

Answer

The Human TNFsR I ELISA Kit PicoKine endotoxin level is <0.5EU/mg antibody and yes it is azide free. Costumers needs to optimize the conditions, as the culture condition is different. The assay was performed on human peripheral blood mononuclear cells, incubated with 10 ng/ml TNSF4 for 5-6 days in serum-containing media. Other cells were incubated with TNSF4 that has been first incubated with 5-15ng/ml at 37C for one hour before adding to the medium.

Question

Q: what is the protocol regarding preparation of cell lysates prepared for use in Picokine® ELISA kits?

Verified Customer

Verified customer

Asked: 2019-12-10

Answer

A: for those Picokine® ELISAs where cell or tissue lysate is a validated sample type, sample preparation instructions for lysate can be found in the product insert. Components in lysate and lysis buffer can effect immunoreactivity, so if lysate is not a validated sample type, care must be taken in sample preparation and validation.

Question

Can EK0528 be used for cell lysate samples? <br>Keywords: sample types

Verified Customer

Verified customer

Asked: 2019-10-08

Answer

EK0528 is used to detect soluble fragment. Our lab hasn't tested EK0528 with any cell lysate, so we do not recommend using EK0528 to test cell lysate samples.

Question

Q: how much samples can be assayed in a Picokine® ELISA Kit?

Verified Customer

Verified customer

Asked: 2019-08-16

Answer

A: The Picokine® ELISA Kits will generally run a 7-point standard curve, non-specific binding wells, and 39 samples in duplicate. this may might differ by kit so please refer to each datasheet for details.

Question

Q: What is the optimal O.D. value for TNF RI ELISA kit? I performed your TNF RI ELISA on serum samples. For my positive control, I received an O.D. value of 0.826, while my negative control received a value of 0.136. I obtained both of these controls from the ELISA kit, where your kit's typical data shows O.D. values much higher than my positive control and your background is lower. My samples O.D. values are around 0.225 and the highest is only 0.357. can I consider these samples contain TNF RI even though the O.D. values are not very high?

C. Williams

Verified customer

Asked: 2019-08-05

Answer

A: The absolute O.D. values may change according to incubation time. The more you incubate the higher the O.D. values are going to be. what you should focus on is whether your sample O.D. values are statistically significantly higher than your blank values. considering your assay, you could extend your development time in the substrate incubation step to obtain higher O.D. values, as long as your negative controls' O.D. values are not increasing faster in proportion to your positive controls. normally, a sample with O.D. value 2 standard deviations higher than your negative controls can be considered positive. We calculate the sensitivity of this ELISA kit by converting cutoff O.D. value, calculated as the average of 20 negative controls plus 2 standard deviations of the 20 negative controls, into a concentration. in other words, when we claim this TNF RI ELISA kit to have sensitivity of 1pg/ml, that means the minimum amount of TNF RI that can be declared/interpreted as positive by the above standard is 1pg/ml.

Question

Q: if the enzyme conjugated TNF RI antibodies are mixed with the substrate, will that convert the substrate into the enzymatic reaction product? Or the enzyme function is only activated when the antibody is attached with the TNF RI antigen?

Z. Collins

Verified customer

Asked: 2018-07-19

Answer

A: The enzyme is always active. Avoid contaminating the substrate with enzyme prior to the incubation otherwise it compromises the assay with false positive signal.

Question

Q: Are Boster Bio recombinant proteins and antibodies sterile?

Verified Customer

Verified customer

Asked: 2018-03-12

Answer

A: although the vials are bottled using aseptic techniques, heat-treated vials, and sterile stock solutions, they are not considered or guaranteed to be sterile. If sterile material is required for an experiment, the material can be filtered through a 0.2 micron filter designed for use with biological fluids.

Question

Q: can I use heparin plasma as samples in Human TNF RI Picokine® ELISA Kit (Catalog # EK0528)?

Verified Customer

Verified customer

Asked: 2015-01-26

Answer

A: Chelating agents such as EDTA, Heparin and Citrate can bind metal ions from the functional domain of TNF RI causing degradation of its protein structure. TNF RI may be denatured as a result and may compromise the assay's measurements. The chilating sites could also be too close to the epitopes required for detection and block the antigen antibody reaction. We have tested the TNF RI ELISA, treating samples with a number of anticoagulants and decided that heparin, EDTA or citrate can be used for treatment of blood/plasma samples. Do not use other anticoagulents when collecting samples.
