Background of B7-1/CD80

The protein CD80 (Cluster of Differentiation 80) is a molecule found on activated B cells and monocytes which provides a costimulatory signal necessary for T cell activation and survival. It is also known as B7.1. The cDNA for B7-1 predicts a type I membrane protein, i.e., one synthesized with a signal peptide that is cleaved upon translocation across the endoplasmic membrane. The protein is predicted to contain 2 extracellular domains structurally similar to those of Ig, a hydrophobic transmembrane region, and a short cytoplasmic domain. The CD80 and CD86 (601020) genes encode B7-1 and B7-2, respectively, which are structurally similar members of the immunoglobulin superfamily expressed on a variety of hematopoietic cell types. stated that B7-1 and B7-2 provide a costimulatory signal to T cells by interacting with CD28 and CTLA4. The standard product used in this kit is recombinant B7-1, D37—K245, which is composed of two single chains acids with the dipolymer.

A: do not freeze and thaw whole blood. erythrocytes are fragile and, if frozen and thawed, will undergo hemolysis rendering the samples useless. To keep your blood samples to test CD80 for a later time, you should let the blood clot in glass tubes and separate the serum to freeze for later analysis.

A: for those Picokine® ELISAs where cell or tissue lysate is a validated sample type, sample preparation instructions for lysate are included in the product insert. Components in lysate and lysis buffer may impact immunoreactivity, so if lysate is not a validated sample type, care must be taken in sample preparation and validation.

A: Boster Bio does not evaluate sample stability. Variations in sample collection, processing, and storage may affect the stabilityof samples. It is recommend to assay sample right after collection when possible, or aliquot into single use volumes and store samples frozen. avoid repeat freeze-thaw cycles with the stored samples in order to prevent protein degradation.
