Background of Il23a
Interleukin-23 subunit alpha (IL-23) is a protein that in humans is encoded by the IL23A gene. IL-23 is a heterodimeric cytokine consisting of two subunits, one called p40, which is shared with another cytokine, IL-12, and another called p19 (the IL-23 alpha subunit). IL-23 is an important part of the inflammatory response against infection. It promotes upregulation of the matrix metalloprotease MMP9, increases angiogenesis and reduces CD8+ T-cell infiltration. Recently, IL-23 has been implicated in the development of cancerous tumors. he International Radiation Hybrid Mapping Consortium mapped the p19 gene to chromosome 12 (stSG47812).

It is recommend to assay sample on-stat collection when possible, or aliquot into single use volumes and store samples frozen. limit repetitive freeze-thaw cycles with the stored samples in order to prevent protein degradation.

A: Chelating agents such as EDTA, Heparin and Citrate can attach metal ions from the functional domain of IL-23A causing disruption of its protein structure. IL-23A may be denatured as a result and may compromise the assay's measurements. The chilating sites could also be too close to the epitopes a must for detection and block the antigen antibody reaction. We have tested the IL-23A ELISA, treating samples with a number of anticoagulants and decided that heparin can be used for treatment of blood/plasma samples. Do not use other anticoagulents when collecting samples.
