Background of Mettl23
Recombination activating gene 1 also known as RAG-1 is a protein that in humans is encoded by the RAG1 gene. The protein encoded by this gene is involved in activation of immunoglobulin V-D-J recombination. The encoded protein is involved in recognition of the DNA substrate, but stable binding and cleavage activity also requires RAG2. Defects in this gene can be the cause of several diseases.
Antibody Validation
Boster validates all antibodies on WB, IHC, ICC, Immunofluorescence, and ELISA with known positive control and negative samples to ensure specificity and high affinity, including thorough antibody incubations.

Figure 1. Western blot analysis of METTL23 using anti-METTL23 antibody (DZ41142).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30ug of sample under reducing conditions.
Lane 1: human HCT whole cell lysates,
Lane 2: human Caco-2 whole cell lysates,
Lane 3: human SHY whole cell lysates,
Lane 4: human U87 whole cell lysates,
Lane 5: mouse GL261 whole cell lysates.
After Electrophoresis, proteins were transferred to a PVDF membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-METTL23 antigen affinity purified polyclonal antibody (Catalog # DZ41142) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and detected by Licor IR800 rabbit antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system.
